In een zuidelijke havenstad speelt zich de roeping van Mattheüs af, een verhaal uit de Bijbel. Jezus staat bovenaan de trappen en wijst naar Mattheüs, om hem als volgeling tot zich te roepen. Mattheüs deinst geschrokken achteruit. In het boek op Mattheüs’ schoot, staan de namen van de makers van het schilderij: Berchem en Weenix. Waarschijnlijk zette Weenix de voorstelling op en schilderde Berchem de groep rond Jezus. Dat voor dit schilderij twee kunstenaars van naam samenwerkten, maakte het tot een begeerd verzamelobject.
